IgA Nephropathy (IgAN)

IgA Nephropathy (IgAN), also known as Berger’s Disease is an autoantibody mediated kidney disease. In people with IgAN, the body, makes an abnormal form of a protein called immunoglobulin A (IgA). While, IgA is a normal part of the immune system, in people with IgAN, the body’s immune system forms autoantibodies against the abnormal form. These autoantibodies and the immunoglobulin proteins form immune complexes, which build up in the glomeruli – the tiny filters in the kidney – causing inflammation and scarring, which impairs how waste is filtered in the kidneys and can lead to chronic kidney disease or kidney failure.

IgAN symptoms include blood in the urine, protein in the urine, high blood pressure, and swelling (edema).

The management of IgAN is primarily focused on reducing proteinuria as this has been definitively correlated to reduce the risk of kidney failure and improve outcomes.

The unmet need in IgAN is high and there is a well understood need for treatments that would prevent the production of the abnormal IgA [and associated autoantibodies??].

How CLYM116 may be able to help

APRIL signally promotes survival of plasma cells, the source of many autoantibodies. It also regulates the process in which some B cells begin to express IgA. In IgAn, blocking APRIL signaling may prevents the production of the abnormal IgA and the resulting autoantibody production and immune complex formation that leads to kidney damage.

Through it’s anti-APRIL mechanism of action, CLYM116 has the potential to reduce IgA and be a disease modifying treatment for IgAN.
